A 52-week, randomized, double-blind, double-dummy, parallel-group, multi-centre, non-inferiority study to investigate the efficacy and safety of depemokimab compared with mepolizumab in adults with relapsing or refractory Eosinophilic Granulomatosis with Polyangiitis (EGPA) receiving standard of care (SoC) therapy
Eosinophilic Granulomatosis with Polyangiitis (EGPA) is a rare condition that causes swelling in small blood vessels, often affecting the lungs, sinuses, and other organs. While standard treatments such as steroid tablets can help, the illness often returns, and steroids can cause unpleasant long-term side effects. This 52-week study compares an experimental medicine, depemokimab, with an approved treatment, mepolizumab. Both medicines aim to calm the immune system by targeting specific white blood cells that cause inflammation. Researchers want to find out if depemokimab is at least as effective as mepolizumab at keeping EGPA in remission (quiet and symptom-free). The study also tracks whether patients can safely reduce their oral steroid dose to a very low level. Taking part helps doctors learn if depemokimab could provide an effective, long-lasting treatment option for people living with this challenging condition.

על מה המחקר הזה?
Eosinophilic Granulomatosis with Polyangiitis (EGPA) is an uncommon condition where blood vessels become inflamed. This inflammation is often driven by a type of white blood cell known as an eosinophil. People with EGPA often experience breathing difficulties, sinus problems, nerve tingling, or skin rashes. For many people, the condition is 'relapsing' or 'refractory', which means the symptoms keep returning or are difficult to bring under control even with standard medical therapy.
Currently, many patients rely on regular steroid tablets (such as prednisolone) and newer biologic medicines like mepolizumab to keep the condition quiet. While steroids are effective at damping down inflammation quickly, using them for long periods can lead to significant problems, such as weakened bones, weight gain, high blood pressure, and a higher risk of infections. Finding treatments that control the disease while allowing steroid doses to be cut down is a major priority for doctors and patients alike.
This trial investigates a newer medicine called depemokimab to see if it works just as well as mepolizumab. Both medicines work by blocking the signals that cause eosinophils to multiply and trigger inflammation. Depemokimab is designed to last longer in the body, which could mean fewer injections over time if it proves successful. By comparing the two treatments directly over one full year, researchers hope to confirm whether depemokimab is a safe, effective option that can help keep EGPA in check and reduce the need for daily steroids.
נקודות עיקריות
- Compares a new medicine (depemokimab) with an existing treatment (mepolizumab) for EGPA.
- Aimed at adults aged 18 and older whose EGPA has returned or is hard to treat.
- The study lasts for 52 weeks and involves regular clinic visits and health checks.
- Key goals are keeping symptoms in remission and safely lowering daily steroid doses.
- Every participant receives active treatment alongside dummy injections to keep the test fair.
- Joining is entirely voluntary and does not impact your normal NHS treatment rights.
מי עשוי להיות זכאי?
This study is open to adults aged 18 and older who have a confirmed diagnosis of EGPA. To take part, your condition must be considered 'relapsing' (meaning your symptoms have returned after previous treatment) or 'refractory' (meaning your symptoms have not fully settled despite your current medication).
You will also need to be taking standard medical treatment for your EGPA, which generally includes a daily dose of steroid tablets. Both men and women may join the study.
Before taking part, the trial doctor will review your full medical history and perform safety checks. You may not be able to join if you have certain other serious health conditions, if you are pregnant or breastfeeding, or if you have had a severe allergic reaction to similar biologic medications in the past.
האם מחקר זה יכול להתאים לך?
ענה על השאלות המהירות הללו כדי לבדוק אם אתה עשוי להיות זכאי. זהו מדריך בלבד — צוות המחקר מקבל את ההחלטה הסופית.
- I am aged 18 or older.
- I have been diagnosed with Eosinophilic Granulomatosis with Polyangiitis (EGPA).
- My EGPA symptoms have flared up recently or have not settled with my current medication.
- I am currently receiving standard treatment, such as daily oral steroid tablets.
- I am able and willing to attend regular hospital appointments for 52 weeks.
מה כרוך בהשתתפות?
Taking part in this study lasts for 52 weeks (around one year). You will be randomly placed into one of two treatment groups by a computer. One group receives depemokimab and a dummy (placebo) version of mepolizumab, while the other group receives mepolizumab and a dummy version of depemokimab. This 'double-dummy' approach ensures that neither you nor your study doctor knows which active drug you are receiving, keeping the results completely fair.
Both medicines are given as injections under the skin. You will visit the hospital or clinic regularly throughout the year. During these visits, doctors will check your symptoms, perform routine blood tests, measure your blood pressure, and assess any side effects. The study team will also work closely with you to see if your daily steroid tablets can be carefully and gradually reduced. When the 52 weeks finish, your doctor will discuss your ongoing care plan with you.

שאלות נפוצות
What does 'non-inferiority' mean in this trial?
It means the study is designed to show that the new medicine, depemokimab, works at least as well as the current standard treatment, mepolizumab, rather than needing to prove it is superior.
Will I receive a placebo (dummy treatment)?
Every participant will receive an active medicine. Because both medicines look different, you will receive injections for both, where one is active and the other is an inactive dummy (placebo). This ensures everyone gets treatment while keeping the study unbiased.
Will I be taken off my regular steroid tablets immediately?
No. You will continue taking your standard care medicines. The medical team will only attempt to lower your daily steroid dose slowly and carefully if your symptoms remain well-controlled.
What does remission mean in this study?
Remission means that your EGPA is completely inactive (no signs of disease activity) and you only need a very low dose of daily steroids (4 mg or less) to stay well.
Do I have to pay to take part in the trial?
No. The trial medication, clinic visits, and all study-related tests are provided free of charge, and travel expenses are often reimbursed.
